چکیده

We derived an ordinary differential equation model to capture the disease dynamics during blood-stage malaria. The model was directly derived from an earlier age structured partial differential equation model. The original model was simplified due to experimental constraint. Here we calibrated the simplified model with experimental data using multiple objective genetic algorithm. Through the calibration process, we quantified the removal of healthy red blood cells and the the preferential infection of reticulocytes during Plamodium cynomolgi infection of Macaca mulatta. The calibration of our model also revealed the existence of host erythropoietic response prior to blood stage infection.

Detection of new Babesia microti-like parasites in a rhesus monkey (Macaca mulatta) with a suppressed Plasmodium cynomolgi infection.

A new type of piroplasm, phylogenetically closest to Babesia microti-like parasites previously detected in Eurasian red squirrels (Sciurus vulgaris orientis), was identified in a rhesus monkey (Macaca mulatta) imported from China. After challenge with Plasmodium cynomolgi M strain blood-stage parasites, the rhesus monkey repeatedly showed markedly reduced levels of Plasmodium parasitemia when c...

Natural killer activity against human K562 tumour cells during Plasmodium cynomolgi malarial infection of rhesus monkeys.

This study assessed the natural killer (NK) cell activity profile during Plasmodium cynomolgi infection in rhesus monkeys. There was a significant decrease in the NK cell activity in the peripheral blood leukocytes of infected monkeys during the early, ascending phase of infection. However, as the parasite load decreased, NK cell activity returned to normal levels. This could be correlated with...

Correlates of severity of disease in Macaca mulatta infected with Plasmodium cynomolgi

Characterization of host responses associated with severe malaria through an integrative approach is necessary to understand the dynamics of a Plasmodium cynomolgi infection. In this study, we conducted temporal immune profiling, cytokine profiling and transcriptomic analysis of five Macaca mulatta infected with P. cynomolgi. This experiment resulted in two severe infections, and two mild infec...

Invasion characteristics of a Plasmodium knowlesi line newly isolated from a human

Plasmodium knowlesi is extensively used as an important malaria model and is now recognized as an important cause of human malaria in Malaysia. The strains of P. knowlesi currently used for research were isolated many decades ago, raising concerns that they might no longer be representative of contemporary parasite populations. We derived a new P. knowlesi line (University Malaya line, UM01), f...

Statistical model to evaluate in vivo activities of antimalarial drugs in a Plasmodium cynomolgi-macaque model for Plasmodium vivax malaria.

Preclinical animal models informing antimalarial drug development are scarce. We have used asexual erythrocytic Plasmodium cynomolgi infections of rhesus macaques to model Plasmodium vivax during preclinical development of compounds targeting parasite phospholipid synthesis. Using this malaria model, we accumulated data confirming highly reproducible infection patterns, with self-curing parasit...
